Abstract

Tuntun Angin (Elaeocarpus floribundus) is one of typical plant of flood exposure. The purpose of this study was to obtain the optimal annealing temperature of 28S rRNA primer in Tuntun Angin DNA for PCR. Reseacrh procedures included sampling, total DNA isolation, electrophoresis, and PCR. In this study, Tuntun Angin leaves collected from Kajuik Lake, Langgam District, Pelalawan Regency, Riau Province were used. The result showed that the length of target band was 600 bp. Optimal annealing temperature for 28S rRNA gene amplification in Tuntun Angin was 50.400C (Tm-5). The band produced was thick, clear, and not smeared.
