Abstract
Laccase is an enzyme that requires additional chemical components (non-protein)
such as metal ions to increase its activity. This study aims to determine the effect
of adding Cu2+, Mg2+ and Zn2+ metal ions and organic solvents acetonitrile,
acetone, ethanol and methanol on the activity of the laccase enzyme produced
from the fungus Trichoderma asperellum LBKURCC1 using ABTS as a
substrate. The concentration of metal ions and organic solvents added were
respectively (1, 5 and 10 mM) and (5, 20 and 40 %). Laccase enzyme activity test
was measured using a UV-Vis spectrophotometer with a wavelength of 420 nm
and incubated for 5 minutes. The highest laccase activity was obtained by adding
Cu2+ metal ions at a concentration of 10 mM with a value of 544.00 ± 12.56 U/L,
which increased laccase activity by 63,43% which acts as a cofactor. Meanwhile,
the addition of organic solvents decreased its activity by 80,17% with a value of
66.00 ± 4.09 U/L in acetonitrile solvent with a concentration of 40% which caused
the enzymes to denature.
