dc.description.abstractDurik-durik plant is categorized as a member of Syzygium genus based on DNA barcoding analysis using intergenic spacer psbA-trnH, matK, rbcl, ITS and trnL-F intergenic spacer. Polymerase Chain Reaction (PCR) is an in vitro DNA amplification technique using a thermal cycler machine to amplify DNA fragments in a relatively short time. PCR process is influenced by the PCR components and the annealing temperature. This research was conducted to optimize annealing temperature of 748F/1318R primer pair. Temperature optimization is analyzed in various annealing temperature. Result showed that the optimum temperature for amplification of ndhF gene is 46.6°C and the amplificon obtained was 600 bp.en_US
