Abstract:
Pandan (Benstonea sp.) from Riau is one of the plants that has an important
role in the Kajuik Lake ecosystem located in Kampar River, Langgam, Pelalawan
Regency, Riau Province. The PCR technique is the technique that used to amplify
DNA in vitro in short time. The process of amplify DNA can occur by the primer.
The purpose in this study was to obtain the optimal annealing temperature for
amplifying trnV-ndhC intergenic spacer on pandan from Riau. In this study, a primer
pair used was trnV(UAC)x2-F dan ndhC-R. The result showed that annealing
temperature for amplifying trnV-ndhC intergenic spacer region was 51,2ºC and
48,2ºC. The PCR product was about 1000 bp.
