Item IDENTIFIKASI PARASIT Clinostomum sp. (TREMATODA) PADA IKAN SEPAT (Trichogaster trichopterus) DENGAN SCANNING ELECTRON MICROSCOPE(2013-05-30) Riauwaty, Morina; WindartiA study on identification of Clinostomum infections in Trichogaster trichopterus in Pekanbaru has been conducted on August to October 2011. This study aims to understand the topography of Clinostomum in Trichogaster trichopterus. Excysted Clinostomum sp. was found in cavity organ were aseptically taken using needle, preserved in absolute ethanol. Morphological examination was done with Scanning Electron Microscopy (SEM). Result shown that topography of the tegument of Clinostomum sp. metacercariae infections in Trichogaster trichopterus in Pekanbaru indicated different structure in individual surface. Oral sucker located at the subterminal end, ellips and was surrounded by fine surface pucker. The ventral sucker was close to oral sucker toward anterior area, and had round sensory papillae without spinae. The body surface was concave without spinae and was completed with round papillae. Round and pucker-surrounded excretory pore appeared at the posterior terminal. Results of Scanning Electron Microscopy indicates that Clinostomum sp. identified in this study was similar to Clinostomum cutaneum.Item A NEW SPECIES OF CLINOSTOMUM (DIGENEA: CLINOSTOMIDAE) FROM CLIMBING PERCH (Trichogaster trichopterus) IN RIAU, INDONESIA(2013-05-30) Riauwaty, Morina; Kurniasih; Prastowo, Joko; WindartiThe aim of study is to identify the Clinostomum sp.obtained from visceral organs of Climbing perch (Trichogaster trichopterus) in Riau, Indonesia. In September 2010, Climbing perch (Trichogaster trichopterus) in Riau, Indonesia were collected, identified and examined for Clinostomum infection. Digenea, encysted metacercariae of Clinostomum were excysted with a needle, fixed in 10% formalin solution for morphological examination. Some samples were fixed with glutaraldehyde for SEM, and with ethanol absolute solution for molecular analysis. The result showed that the metacercariae of Clinostomum sp. from Riau had a body shaped like leaf. The surface of the body had not spinae. The anterior part was narrower than posterior. Posterior end was round. Body length had size of 347.2 (298.6 to 395.8) μm, body width of 127.2 (115.7 to 138.6) μm. Oral sucker small, terminal and had size of 16.2 (14,29 to 18,6) μm. Ventral sucker larger than oral sucker and had size of 63,6 μm. Clinostomum sp. has similarities to Clinostomum phalacrocorasis based on molecular study. This research was the first investigation of Clinostomum sp. in Climbing perch (Trichogaster trichopterus) as second intermediate host found in Riau, Indonesia.Item PEMANFAATAN BAHAN ALAMI UNTUK MENCEGAH DAN MENGOBATI PENYAKIT IKAN EKONOMIS PENTING RIAU(2013-05-28) Riauwaty, Morina; Lukistyowati, IesjeA study on histopathology of liver and kidney of Pangasius hypopthalmus that is infected with Aeromonas hydrophila and treated with Curcuma xanthorrhiza Roxb has been conducted on March-July 2012. This study aims to understand the histological structure of the liver and kidney of the fish that is treated with C. xanthorrhiza extract. There were 3 treatments applied. The treated fishes were infected with A. hydrophila (0.1 ml of 109 of A. hydrophila culture) and then were immerse in 3 different concentrations of C. xanthorrhiza extract, they were T1 (0.2 g/l); T2 (0.4 g/l) and T3 (0.6 g/l) for 5 minutes/ day for a 2 weeks period. The negative control were fishes that were not receive any treatment, while the positive control were fishes that were infected with A. hydrophila, and were not treated with C. xanthorrhiza extract. The liver showed vacuolar degeneration in the hepatocyte, necrosis, hemorrhage and hipertrophy in the sinusoid. The kidney showed vacuolar degeneration in the epithelium of renal tubulus, necrosis, haemorrhage and dilation in Bowman’s capsule. Based on data obtained, it can be concluded that immersion of fish in C. xanthorrhiza extract is able to cure A. hydrophila infection on fishItem PENERAPAN MODEL PEMBELAJARAN LANGSUNG MELALUI PHYSICAL SELF ASESSMENT UNTUK MENINGKATKAN AKTIVITAS DAN HASIL BELAJAR MAHASISWA FAPERIKA PADA MATA KULIAH MANAJEMEN KESEHATAN IKAN(2013-05-28) Riauwaty, Morina; Lukistyowati, IesjePenelitian tindakan kelas ini bertujuan untuk meningkatkan aktivitas dan hasil belajar mahasiswa FAPERIKA pada mata kuliah Manajemen Kesehatan Ikan dengan penerapan model pembelajaran langsung kooperatif melalui physical self asessment. Penelitian ini dilaksanakan di Jurusan Budidaya Perairan Fakultas Perikanan dan Ilmu Kelautan Universitas Riau pada semester Genap Tahun Ajaran 2011/2012. Subjek penelitian adalah mahasiswa Jurusan Budidaya Perairan yang mengambil mata Manajemen Kesehatan Ikan yang berjumlah 54 orang (24 perempuan dan 20 laki-laki). Metode pembelajaran langsung (direct instruction) dirancang khusus untuk menunjang proses belajar mahasiswa yang berkaitan dengan pengetahuan deklaratif dan pengetahuan prosedural yang terstruktur dan dengan pola yang bertahap. Parameter penelitian yang diamati adalah keterampilan proses, hasil belajar mahasiswa, aktivitas belajar mahasiswa dan aktivitas dosen. Prosedur penelitian meliputi, tahap persiapan, tahap pelaksanaan, observasi dan refleksi. Data keterampilan proses, hasil belajar (daya serap, ketuntasan belajar mahasiswa), aktivitas mahasiswa dan aktivitas dosen akan dihitung berdasarkan (Purwanto, 2007) dan dianalisa secara deskriptif. Hasil penelitian tindakan kelas menunjukkan bahwa hasil daya serap mahasiswa mengalami peningkatan dari dari 75,44% (cukup) pada siklus I dan menjadi baik (85,55%) pada siklus II, ketuntasan mahasiswa juga mengalami peningkatan dari 81,04% pada siklus I dan 87,93% pada siklus II, aktivitas mahasiswa mengalami peningkatan dari kategori cukup (83,61%) pada siklus I, menjadi kategori baik (90,42%) pada siklus II. Dari hasil ioni dapat disimpulkan bahwa model pembelajaran langsung melalui physical self assesment dapat meningkatkan aktivitas dan hasil belajar mahasiswa pada mata kuliah Manajemen Kesehatan Ikan.Item PREVALENSI CLINOSTOMUM (DIGENEA, CLINOSTOMIDAE) PADA IKAN SEPAT RAWA (Trichogaster trichopterus) DARI RIAU, INDONESIA(2013-05-30) Riauwaty, Morina; KurniasihThe aim of study was to know the prevalence of Clinostomum infections in Climbing perch (Trichogaster trichopterus) in Riau, Indonesia. Trichogaster trichopterus from Sail river, Riau were collected, and examined for Clinostomum infection. Encysted metacercariae of Clinostomum were excysted with a needle and fixed in 10% formalin. Speciemens were stained with Semichon’s Acetocarmine, dehydrated in alcohol series, cleared with xylene and mounted in entellan. The Clinostomum parasites were observed under a stereomicroscope and numbers recorded. Clinostomum identification was based on Chung et al 1995. The result showed that 18 of 105 observed climbing perch were infected by Clinostomum sp. The infected fish had total length between 5,6-11cm, while the weight range was 2,01-28,64 g. Based on the sexuality, the male fish was infected by the Clinostomum sp. (76,2%) more than the female (23,8%). Prevalence of Clinostomum sp. infection to the fish was 17,1%, and the infestation was between 1 to 29 worm per location. Freshwater fish were collected from Sail River, identified and examined for morphological observation. Encysted metacercariae of Clinostomum were excysted with a needle, fixed in 10% formalin and stained with Semichon’s Acetocarmine. Clinostomum parasites were observed under a stereomicroscope and numbers recorded. The result showed that infected freshwater fish were identified as Trichogaster trichopterus and prevalence of Clinostomum infection to the fish was17,1%. The infected fish had total length between 5,6-11cm, while the weight range was 2,01-28,64 g. The cysts were 1,5-2 mm in diameter, with a thin (c. 0,9 μm wide). There was no spinae in tegument dan had 2 suckers (oral and ventral), posteriorly located genitalia and paired digestive caeca. This digeneans were identified as the metacercariae of Clinostomum sp. Parasit ini bersifat zoonotik yang menyebabkan penyakit “laryngo-pharingitis” dan dapat mengakibatkan kematian karena terjadi Asphyxia pada manusia (Kifune dan Kousaka, 1994; Vianna et al., 2005). Penularan pada manusia dapat terjadi bila memakan ikan mentah yang terinfeksi Clinostomum complanatum atau memakan ikan yang tidak dimasak secara sempurna. Kasus ini terjadi pertama sekali di Jepang (Yamashita, 1938; Kifune et al., 2000), Israel (Witenberg, 1944), India (Cameron, 1945), Korea (Chung et al., 1995a), Thailand (Chung et al., 1995a; Tiewchaloern et al., 1999) dan Iran (Kifune et al., 2000). Infeksi Clinostomum complanatum pada ikan air tawar di Indonesia mengakibatkan kematian ikan, kegagalan dalam usaha budidaya ikan dan kerugian ekonomi (Mitchell, 1995). Ikan yang terinfeksi parasit ini menunjukkan gejala perubahan tingkah laku, pertumbuhan ikan terganggu, berat badan menurun, kehilangan napsu makan, frekuensi berenang berkurang, warna tubuh pucat, pergerakannya menjadi lambat (Lacerda et al., 2008; Mwita dan Nkwengulila, 2008). Ikan akan mati jika ditrasportasikan dalam jarak jauh (Lo et al.,1982). Akibat adanya kasus ini, maka penelitian tentang morfologi dan molekuler Clinostomum complanatum terus berkembang. Selama ini informasi tentang identifikasi yang akurat untuk mendiagnosa suatu penyakit dan menanggulangi penyebab penyakit di Indonesia masih terbatas, oleh sebab itu penemuan spesies baru pada suatu daerah dapat digunakan sebagai alat diagnostik suatu penyakit. Berdasarkan uraian tersebut maka penelitian tentang studi morfologi dan molekuler Clinostomum complanatum di Yogyakarta perlu dilakukan sebagai data awal untuk mengetahui kekerabatan parasit di Indonesia.
