Abstract:
Nona makan sirih (Clerodenrum thomsoniae) is a medicinal plant from the family
Lamiaceae, and the genus Clerodendrum. It is usually grows as an ornamental plant.
This study aimed to isolate and characterize the pure compound from the n-hexane
extract of the nona makan sirih leaf plant (Clerodendrum thomsoniae) and to analyze a
its toxicity of the isolated compound and n-hexane extract of the nona makan sirih leaf
(Clerodendrum thomsoniae) using the brine shrimp lethality test (BSLT) method. Nona
makan sirih was macerated using methanol as a solvent and then fractionation using
n-hexane. The separation of compounds was carried out by vacuum liquid
chromatography (VLC) and obtained 12 fractions. In fraction 2, there was one spot on
the TLC plate which indicated this fraction was pure and coded as CT-H2. The purity
test of CT-H2 was analyzed by thin layer chromatography test (TLC) with several
eluents mixtures. CT-H2 were characterized using a FT-IR spectrophotometer. The
results of the phytochemical characterization and analysis of the CT-H2 compound
showed some characters of terpenoid. Toxicity test showed the LC50 values of CT-H2
and n-hexane fraction were 254.22 μg/mL and 209.79 μg/mL, respectively. The LC50
value <1000 μg/mL indicates that the n-hexane fraction and CT-H2 have potential
activity be developed as a cytotoxic agent.
