Item EVALUASI KARAKTER FENOTIPIK DAN UPAYA PENGEMBANGAN TANAMAN PADI LOKAL MELALUI PERSILANGAN DAN MUTASI BUATAN(2014-03-05) Wahibah, Ninik Nihayatul; Herman; Roslim, Dewi IndriyaniPadi (Oryza sdtiva L.) rnerupakan sumber pangan bagi masyarakat Indonesia yang dikonsumsi tidak kuran g dari 200 juta penduduk. Konsumsi beras rata-rata 133 kglkapita/tahun, maka total kebutuhan beras 26,6 juta ton/tahun (Husodo 2007). Pertarnbahan jurnlah penduduk nrendorong rneningkatnya kebutuhan akan beras, oleh karena itu perlu digalakkan usaha peningkatan produksi beras untuk memenuhi kebutuhan tersebut. Angka Tetap (ATAP) prodtrksi padi tahun 2010 sebesar 574.864 ton padi Gabah Kering Giling (GKG) atau meningkat dibanding produksi padi pada tahun 2005 sebesat 425.095 toMra (BPS 2011). Peningkatan produksi padi tersebut belnm mencuktrpi kebutuhan konsumsi beras penduduk R.iau sebesar 659.6rc tonlha. Hal ini menyebabkan tingginya ketergantungan pasokan beras dari Provinsi lain seperti Sumatera Barat atau Sumatera Utara. Untuk mengurangi ketergantuttgaf tersebut, pemerintah Provinsi beserta pelnerintah Kabupaten se Riau mengeluarkan kebijakan peningkatan produksi beras. Kebiikan tersebut dapat diwujudkan dalam bentuk progrimr ekstensifikasi manpun intensifikasi. Upaya ek-stensiJikasl dilakukan dengan rnengernbangkan areal pertanian seperti mengembangkan varietas padi pada lahan gambut yang barryak terdapat di Provinsi Riau. Information related to genetic resources of castor plant is little known. Molecular approaches can be used to analyze variation of germplasm. Molecular-based analysis is strongly influenced by quantity and purity of DNA sample as template. Therefore, this research aimed to isolate genomic DNA of castor bean using Genomic DNA Mini Kit (Plant) GeneAid. The result showed that extraction DNA total produced DNA bands . Five individuals show thin fragmen and six individuals show thick fragment.Item ISOLASI DNA GELAM (Melaleuca leucadendra Linn.) MENGGUNAKAN GENOMIC DNA MINI KIT (PLANT) GENEAID(perpustakaan UR, 2021-07) Nurafni, Indah; Wahibah, Ninik NihayatulGelam (Melaleuca leucadendra Linn.) belongs to Myrtacea family potentially producing both wood and non-wood products. However, gelam germ plasm is still underutilized leading to be threatened by their extinction. Natural population of gelam can be found in peat area in Kepulauan Meranti, Riau. Molecular genetic analysis of gelam germ plasm may facilitate to genetic conservation and genetic improvement of the population. DNA isolation is the most important step in the molecular-based analysis. Therefore, we aimed to isolate genomic DNA using DNA extraction kit (Genomic DNA Mini Kit (Plant) Geneaid). Result of the study showed that genomic DNA successfully extracted from gelam young leaves. It indicates that DNA extraction kit is powerful to provide DNA template for further analysisItem KEANEKARAGAMAN GENETIK MANGGIS (Garcinia mangostana L.) PULAU BENGKALIS MENGGUNAKAN PENANDA ISSR(2014-03-04) Fitmawati; Wahibah, Ninik Nihayatul; Aryantri, RisaManggis (Garcinia mangostana L.) memiliki nilai ekonomi tinggi dan saat ini menempati urutan pertama ekspor buah segar Indonesia dengan penambahan volume sebesar 10,7% per tahun. Sampai saat ini permintaan pasar buah manggis tetap tinggi, sehingga masih berpeluang untuk dikembangkan. Karena itu perlu dilakukan eksplorasi kekayaan plasma nutfah manggis. penelitian keanekaragaman genetik manggis secara molekuler terhadap 13 aksesi kandidat tetua manggis unggul Bengkalis tersebut. Studi genetik tanaman dapat dilakukan melalui analisis genetik kandidat tetua manggis unggul Bengkalis akan dilakukan dengan menggunakan penanda molekuler yaitu ISSR (Restriction Fragment Length Polymorphisme). Amplifikasi DNA dengan 6 primer ISSR menghasilkan 49 pita DNA yang bervariasi (300-1125 pb) terdiri atas 28 pita polimorfik dan 21 pita monomorfik. Hasil penelitian ini menunjukan variabilitas genetik yang cukup tinggi berdasarkan analisis klaster yang membentuk 2 klaster utama dengan koefisien kemiripan berkisar 63-94% dan keanekaragaman genetik 6-37%. Analisis komponen utama berdasarkan pita DNA ISSR dan kombinasi data morfologi dengan molekuler menunjukkan Keanekaragaman yang tinggi 96% dan 90 % pada 49 dan 152 karakter.Item KEANEKARAGAMAN GENETIK RAMIN (Gonystylus bancanus Miq. Kurz) DI CAGAR BIOSFER GIAM SIAK KECIL-BUKIT BATU KABUPATEN BENGKALIS PROVINSI RIAU BERDASARKAN POLA PITA ISOZIM(2014-03-28) Syafitri, Meilian; Wahibah, Ninik Nihayatul; Fatonah, SitiRamin is timber plant with high demand in furniture industries because it has good wood and fiber texture. The high selling prices and market demand has led to increase illegal logging, consequently ramin population will be endangered. Therefore, it is necessary to study the genetic diversity as one of the conservation effort of ramin using biochemical markers. This study was aimed to analyze the genetic diversity of ramin (Gonystylus bancanus Miq. Kurz) in Giam Siak Kecil-Bukit Batu Bengkalis biosphere reserves using an isozymes technique. Isozyme analysis was performed using the horizontal model of starch gel electrophoresis. A total of 50 individual ramins were used for analyzing the isozyme banding patterns using one enzyme namely peroxidase (PER). The result showed clear banding patterns. PER enzyme produced twelve banding patterns and migrated to the anode (positive) and cathode (negative) poles. SAHN clustering analysis using UPGMA method showed that all of the individuals of ramin were clustered with the similarity coefficient of 38% and separated into two groups (I and II). All of the members of first cluster were grouped with the similarity coefficient 61%, while the second cluster members were grouped with similarity coefficient 55%. However, excess Fe is toxic, causing multiple damage to celluler structures. Commonly, tidal wetlands rice variety relative tolerance to waterlogged conditions and excess Fe. Mechanism of plants to tolerate the excess Fe is related to the action of ferritin protein. The objective of this study was to compare partial ferritin gene sequence between riau rice variety with national tidal wetlands rice variety, Siam Sintanur. This research was conducted through DNA extraction with CTAB method, amplification using forward primer OsFer_E5 and reverse primer OsFer_E8, sequencing, sequence analysis using BLAST and ClustalW program. There was a SNP (single nucleotide polymorphism) of partial ferritin gene sequence at base-114. IR64 and Siam Sintanur had base-Adenin while Bakung, Siputih, and Serei had base-Citosin at that position. SRAP (Sequence Related Amplified Polymorphism) marker has been reported more useful for revealing diversity germplasm. Our research aimed to explore genetic diversity of castor bean based on marker SRAP. Genomic DNA was extracted from leaves tissue based on Genomic DNA Mini Kit Plant (Geneaid) protocol. Profile SRAP data was converted to score data and analyzed using NTSYSpc 2.02. The results showed similarity amongst samples based on SRAP band profiles was 0,50–0,87. PIC value (0,25) showed moderate-informative category.
