dc.description.abstract |
Biosphere Reserve GSK-BB is an area of peat ecosystem that has been largely
converted, causing a decrease in the quality of the peat. Generally, the parameters
of peat quality use biological, physical and chemical factors. This parameter has
not shown high sensitivity. Therefore, Bacterial Key Species (BKS) was
developed from 16S rRNA gene sequences as sensitive parameters by DNA
barcode. This study aimed to find the DNA barcode of the detected BKS and the
specific primer design and in silico primer testing. The method of determination
was carried out by selecting sequences based on phylogenetic trees and BLASTn.
Furthermore, the primer design and specificity test were carried out using the in
silico PCR technique. The results of the study detected 5 BKS groups from
secondary forest and HTI (BKS-SHI) and 5 DNA barcode candidates. Candidates
for DNA barcode after primer design and specificity test obtained one DNA
barcode namely barcode DNA BKS-SHI3. Specific primers obtained from the
DNA barcode BKS-SHI3 contained 6 primers, BKS-SHI3-P2, BKS-SHI3-P3,
BKS-SHI3-P5, BKS-SHI3-P7, BKS-SHI3-P8, and BKS-SHI3-P9. The detected
DNA barcode BKS-SHI3 can be used as a peat monitoring tool, after the DNA
barcode has been registered into GenBank. |
