Abstract:
Nibung (Oncosperm tigillarium (Jack) Ridl.) belongs to family Arecaceae. Characterization
based on morphological features in nibung is difficult, therefore molecular characterization is
necessary to be done. Molecular characterization based on DNA markers provides a fairly
high degree of accuracy. Plant DNA isolation is the key step in genetic and molecular
experiments. One factor that has an important effect on DNA isolation is leaf condition. This
research was carried out with the aim to find out the quality of DNA from the isolation of
fresh nibung leaves and dried leaves storing in silica gel. Based on the results of this study,
DNA isolation of the nibung dried leaves showed DNA band with good quality compared to
fresh leaves.
