Abstract

Polyhydroxyalkanoate (PHA) as one of the polymers for making bioplastics had potential to subtitute synthetic plastics that are environmentally unfriendly. By optimization of PHA production using cheap and renewable substrates, production costs can be reduced. In this study, modified palm oil mill effluet (POME) and coconut water enriched media will be used to optimize the production of PHA by soil bacteria from Riau. Cultures of bacteria were purified and selected using Sudan Black B (SBB) techniques, and the selected isolates were used to optimize the production of PHA with three different enriched media, 50% POME, 50% coconut water, and 25% coconut water. Based on the results of bacterial purification and selection, 6 of 19 isolates were highly absorbent. HTI3_3, HS4_1, KS4_1, and TPAS1_5 isolates were selected to optimize production. The results showed the highest dry weight and PHA accumulation were 1,49 g/L and 82,27%, respectively, produced by isolate KS4_1 using 25% coconut water enriched media.
