Abstract:
Cassava (Manihot esculenta Crantz.) is a wellknown plant in the world, especially in tropical countries. In Indonesia, cassava is the third primary food after rice and corn. Province of Riau has several varieties of cassava that have the potential to be developed and they are distributed in several districts such as Kampar and Siak. However, their DNA is not yet analized. The objective of this study was to isolate the total DNA of Roti and Menggalo cassava genotypes. The isolation of total DNA was conducted by taking 200 mg of cassava leaves. The DNA was isolated using modified Cethyl Trimethyl Ammonium Bromide (CTAB) method. The total DNA were then migrated in 1.2% agarose gel to know the quantity and quality of total DNA obtained. The total DNA of Roti and Menggalo genotypes that obtained in this study showed good quality with thick band, intact and not smear. The quantity ranged from 500-667 ng/μl.
