Abstract:
Durik-durik (Syzygium sp.) from Riau is one of the plants that has an important
role in the Kajuik Lake ecosystem located in Kampar River, Langgam, Pelalawan
Regency, Riau Province. The purpose in this study was to obtain the optimal annealing
temperature for amplifying rpl16 intron on durik-durik (Syzygium sp.) from Riau.
Methods included total DNA extraction, electrophoresis, and annealing temperature
optimization. The primer used in this study was 71F and 1516R. The results showed that
the annealing temperature optimization resulted DNA band at temperature 55,3ºC, 53,7ºC
dan 50,2ºC. The next step was to choose the optimal annealing temperature based on the
clear, thick and single band. In conclusion, the optimal annealing temperature for
amplifying rpl16 intron region was 50,2°C. The PCR product was about 1000 bp.
