Item AKTIVITAS ANTIDEPRESAN EKSTRAK ETIL ASETAT BIJI PINANG TUA DENGAN METODA FORCED SWIM TEST PADA MENCIT JANTAN (Mus musculus)(Elfitra, 2023-07) Sumarni, Sumarni; Novianty, RirynDepression is a mental disorder characterized by a persistently lowered mood and leads to patients experiencing decreased productivity and even ending in suicide. Antidepressants can be used for the initial treatment of depressed patients. Herbs has potential as antidepressants that can be selected using the Forced Swim Test (FST) method. This study aims to determine the antidepressant activity of the ethyl acetate extract of old areca nut seeds through the FST method in an acute manner. FST was used because it was one of the most widely used in vivo methods for antidepressant screening in basic and preclinical studies. Intraperitoneal injection of mice was performed 1 hour before FST at a dose of 10 mg/kg and the control group was injected with 0.9% saline only. Ethyl acetate extract of old areca nut seeds dose of 10 mg/kg was not effective as an antidepressant because the decrease in immobility time was not significantly different to control.Item AKTIVITAS ANTIDEPRESAN EKSTRAK METANOL BIJI PINANG MUDA DENGAN METODA FORCED SWIM TEST PADA MENCIT JANTAN (Mus musculus)(Elfitra, 2023-05) Neldi, Mitra Frecilia; Novianty, RirynDepression is a mood disorder in the form of deep depressing emotions or feelings. Depressive disorders often occur in society and become the biggest contributor to suicide. The main pharmacological therapy for depressive disorders is antidepressant drugs. The Forced Swim Test (FST) method is a method commonly used for antidepressant screening. This study aims to determine the antidepressant activity of young areca seed methanol extract through the FST method in an acute manner. The in vivo FST method was chosen because it can be a specific test to measure behavior that is common for antidepressant treatments that have pharmacological effects. The injection was carried out intraperitoneally 1 hour before FST at a dose of 10 mg/kg and the control group was only injected with 0.9% saline. Pharmacological therapy for depressive disorders is a drug from the class of antidepressants. The method commonly used for antidepressant screening is the Forced Swim Test (FST). This study aims to determine the antidepressant activity of n-hexane extract of nutmeg through the FST method in an acute manner. FST is used because it is an in vivo method which can be a special test in basic and pre-clinical research. The injection was carried out intraperitoneally 1 hour before FST at a dose of 50 mg/kg and the control group was only injected with 0.9% saline. The n-hexane extract 50 mg/kg of nutmeg seed had no potential as an antidepressant as evidenced by the decrease in immobility time which was not significantly different from the control.Item AKTIVITAS ANTIDEPRESAN EKSTRAK N-HEKSANA BIJI PINANG MUDA DENGAN METODA FORCED SWIM TEST PADA MENCIT JANTAN (Mus musculus)(Elfitra, 2023-05) Ya’la, Fitra Audhi; Novianty, RirynDepression is a mental disorder caused by an imbalance of neurotransmitters which can be treated with antidepressants. The method commonly used for antidepressant screening which is validated by animal behavior is the Forced Swim Test (FST). It has not been clarified the antidepressant activity of n-hexane extract young areca nut (Areca catechu L.). Therefore, this study aimed to perform FST on mice which had been injected acutely with young areca nut n-hexane extract. The young areca nut n-hexane extract was injected intraperitoneally 1 hour before FST to mice at doses of 100 mg/kg, while the the vehicle control group was only injected 0.9% saline. The n-hexane extract young areca nut seeds at a dose of 100 mg/kg has no potential as an antidepressant. This was evidenced by the unsatisfactory results, the extract induced a significant decline in the immobility time of the mice that had been injected with the drug sample.Item ANALISIS STUDI IN SILICO SENYAWA GUINEENSINE SEBAGAI KANDIDAT OBAT ANTIDEPRESAN(Elfitra, 2022-01) Sirait, Vera Vebrianti; Novianty, RirynDepression is an emotional and mental disorder due to the catalytic activity of the MAO enzyme, causing a neurotransmitter imbalance which is generally treated with antidepressants. Research on antidepressants in vivo has been carried out, but in silico the active compound of Javanese chili (Piper longum BI) has not been studied to see its molecular interactions. Therefore, this study was conducted to inhibit the activity of the MAO-A enzyme (PDB ID: 2Z5X) by administering a guineensine compound using SwissADME to predict pharmacokinetic properties and drugs-likeness properties as well as molecular docking methods using PyRx 0.8 and PyMOL to observe the binding interactions that occur. between the receptor and the ligand. It is known that the guineensine compound does not meet the parameters of pharmacokinetic properties and parameters of drug-likeness properties. Although it is based on a boiled egg that can penetrate the BBB (Blood Brain Barrier), the bioavailability radar shows that guineensine compounds are outside the radar and have a bioavailability score of 0.5. Based on the docking results, it is known that guineensine has a bond free energy value of -4.4 kcal/mol and has the same amino acid residue binding as the native ligand (HRM), namely TYR444 and TYR407 on the active site of MAO-A. So it can be concluded from the results of SwissADME and molecular docking that guineensine compounds are less potential as candidates for antidepressant drugs.Item Mempelajari Daya Inhibisi Ion Cu' Dan Ca' Serta Pola Inhibisinya Terhadap Enzim Akonitase Yang Diisolasi Dari Buah Kalikih Alang (Ricinus Communis)(2015-05-11) Novianty, Riryn; Chaidir, Zulkarnain; Mardiah, ElidaPenelitian tentang inhibisi aktivitas enzim akonitase dengan menggunakan inhibitor ion Cu" dan Ca' telah dilakukan. Enzim akonitase diisolasi dari buah Ricinus communis dengan menggunakan aseton dingin. Untuk menentukan aktivitas dari akonitase digunakan metoda pengukuran substrat asam sitrat sisa berdasarkan metoda LStahre's test. Kadar protein ditentukan dengan menggunakan metoda Lowry. Optimasi dari enzim akonitase didapatkan dari hasil pH optimum 7,3, temperatur optimum 40 °C, dan konsentrasi substrat 1 %. Dari kurva Lineaweaver Burk diperoleh harga Km = 0,104 M. Sedangkan untuk mengetahui daya pola inhibisi ion Cu" dan Ca' dilakukan dengan menvariasikan konsentrasi dari masing-masing ion tersebut. Daya inhibisi ion Cu" dengan konsentrasi 0,1 M adalah 93,347 % dan daya ion Ca' pada konsentrasi 0,1 M adalah 60,124 %. Hasil tersebut menunjukkan pola inhibisi yang sama yakni inhibisi non kompetitif dan masing-masing ion memiliki daya inhibisi yang berbeda.Item OPTIMASI SUHU DAN AGITASI PRODUKSI ENZIM AMILASE DARI JAMUR TERMOFILIK Aspergillus sp. LBKURCC304 through a submerged fermentation process. This study aims to optimize the environmental conditions for the growth of Aspergillus sp. LBKURCC304 in produced amylase enzymes. The results of this study showed that the maximum amylase enzyme production by Aspergillus sp. was obtained at 40ºC and agitation rate of 50 rpm with an amylase enzyme activity of 0.4777±0.0085 U/mL and specific activity of 1.0599± 0.0086 U/mL mg.Item PENENTUAN KURVA PERTUMBUHAN PSEUDOMONAS SP. The purpose of this study was to determine its growth phase Pseudomonas sp. was tested in a Minimal Medium (MM) liquid containing 0,2 mM naphthalene and incubated for 7 days. Optical Density (OD) of isolate were analysed to using spectrophotometer UV-Vis. The result showed that Pseudomonas sp. LBKURCC149 was able to grow and exponential phase occurs from the beginning to the the third day of incubation with optimum OD’s value is 0,104.Item SIMULASI DINAMIKA MOLEKULER SENYAWA KOMPLEKS MOA-A DAN SAFROL SEBAGAI KANDIDAT ANTIDEPRESAN MENGGUNAKAN APLIKASI GROMACS(Elfitra, 2022-01) Ananta, Sony; Novianty, RirynMolecular dynamics (MDS) simulation of the complex compound of nutmeg (Myristica fragrans) with protein is very important to see the molecular interactions that occur, but so far no one has done it. Previous studies in docking and pharmacokinetics have concluded that safrole is an inhibitor that has no potential for MAO-A, but the docking interaction occurs in a static state and without solvent. This study aims to conduct MDS to see the dynamic interaction of the MAO-A complex and safrole in the presence of water as a solvent. MDS can be done using the GROMACS application which is run on the LINUX terminal on UBUNTU and the conformational changes and interactions that occur can be observed with BIOVIA Discovery Studio and PyMOL. The results of the MDS in the form of graph values of root mean square deviation (RMSD), root mean square fluctuation (RMSF), and radius of gyration (Rg) which stated that the stability of the safrole complex was unsatisfactory when compared to native ligands and positive control. Changes in interaction occurred in all test ligands, but safrole did not have the same interaction with amino acid residues at the hydrophobic MAO-A active site as the positive control. The MDS results stated that safrole is a ligand that has no potential as an MAO-A inhibitor obtained from nutmeg based on the stability of the complex and the interaction at the active site, similar to the results of molecular docking under rigid conditions.Item STUDI IN SILICO SENYAWA SAFROLE SEBAGAI KANDIDAT OBAT ANTIDEPRESAN(Elfitra, 2022-01) Maylinda, Shela; Novianty, RirynSince the spread of the COVID-19 pandemic, people's movements have become limited which can affect mental health, one of which is psychological conditions, namely depression. Research on depression has been carried out using in vivo and in vitro methods, but in silico studies related to the active compound of nutmeg seed (Myristica fragrans) have not been carried out. The purpose of this study was to observe the activity of the active compounds in nutmeg seeds (Miristicin, elemicin, safrole and isoeugenol) in inhibiting the action of the MAO-A enzyme. The method used is SwissADME and molecular docking using PyRx 0.8 and PyMOL. The results of prediction of pharmacokinetic properties and drug-likeness using swissADME showed that safrole and positive control compounds can penetrate the BBB (Blood-Brain Barrier) and meet Lipinski's rule, Veber's rule, Ghose's rule and have good skin permeability marked by a bioavailability score of 0.55. . The molecular docking results showed that the positive control compound (moclobemide) and the safrole compound only had 4 amino acid residues in common with the native ligand and 2 of them were active sites. Based on the results of SwissADME analysis and molecular docking, safrole is less potential to be a candidate for antidepressant drugs. Safrole has a bond free energy value of -5.87 kcal/mol which is greater than the positive control and native ligand and based on the interaction of the bonds formed, it shows that safrole can bind to the active site of MAO-A with the similarity of amino acid residues with the native ligand and control. positive.
