dc.description.abstract Siam orange (Citrus nobilis Lour.) from Kampar is one of the main commodity in Riau province which has a sweet taste, aromatic and thin-skinned fruit. Tissue culture was an alternative way in micropropagation of siam orange from Kampar to produce a large number of clone and identical plant. The rooting stage was a very important part to produce planlets in in vitro micropropagation, while the media and auxin consentration played an important role in inducting plant root. The aim of this study was to know the effect of medium type and auxin concentrations on siam orange from Kampar rooting. This study had been conducted in the Laboratory of Integrated Biology, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Riau from July to October 2012, using a randomized block design with factorial treatment. The first factor was the type of medium (half strength and full strength medium). The second factor was the concentration of NAA (0 mg/l, 0,5 mg/l, 1 mg/l, 1,5 mg/l and 2 mg/l). The data was analyzed using Analysis of Variance. The results of this study showed that half strength MS medium was the best medium in rooting induction with the percentage of root growth was 74%. The combination of half strength MS medium with 1 mg/l NAA was the most optimum concentration in rooting induction of siam orange from Kampar, with the percentage of root growth, the earliest rooting day after culturing and the the highest average number of root were 80%, 11 days and 7,3 units respectively en_US
